Exponential Therapeutics is advancing a structured research program focused on evaluating whether controlled, reversible induction of hTERT can be translated into a viable therapeutic strategy. Our development pathway is designed to move from foundational proof-of-concept through progressively more rigorous validation and preclinical evaluation.

Phase 1: Proof of Concept

Current Focus

This initial phase is designed to directly test the core scientific hypothesis: whether hTERT expression can be induced in a controlled, measurable, and reversible manner using small molecules.

Key objectives include:

  • Demonstrating reliable induction of hTERT expression
  • Characterizing dose-response relationships
  • Evaluating reversibility following removal of the inducing agent
  • Assessing early cellular responses and markers of senescence
  • Identifying initial safety signals in controlled systems (including cytotoxicity)
  • Replication of results across multiple cell types

Experimental evaluation in this phase includes direct measurement of telomere dynamics, telomerase activity, and cellular outcomes using established assays. These include qPCR and Q-FISH for telomere length, TRAP assays for telomerase activity, and assessment of senescence and genomic stability markers such as β-galactosidase staining, p16/p21 expression, and γ-H2AX foci.

Studies are designed to evaluate dose-response relationships, reversibility following withdrawal, and longitudinal cellular behavior across multiple passages, providing a structured framework for identifying both beneficial effects and potential risk thresholds.

The goal of this phase is not broad application, but clarity—establishing whether controlled telomerase induction behaves in a predictable and testable manner.

Phase 2: Validation and Expansion

Planned

Following successful proof-of-concept, the program expands into broader validation across additional models and conditions. This phase focuses on strengthening confidence in both efficacy and control.

Key objectives include:

  • Extended evaluation of telomere maintenance over time
  • Further characterization of control parameters (dose, duration, distribution)
  • Exploration of cellular response pathways (e.g. using RNA-seq)
  • Identification of optimal therapeutic windows

This phase is designed to transition from initial signal detection to reproducible and scalable biological effects.

Phase 3: Preclinical Development

Future

With sufficient validation, the program advances toward preclinical development. This phase focuses on translating controlled telomerase induction into a framework suitable for therapeutic development.

Key objectives include:

  • Evaluation in relevant biological models
  • Preliminary pharmacokinetic and pharmacodynamic profiling
  • Safety and toxicity assessment
  • Identification of potential disease applications
  • Preparation for regulatory pathway considerations

This phase represents the transition from exploratory research into structured therapeutic development.
